| Description | Capillary electrophoresis (CE) combined with quadrupole - time of flight mass spectrometer (QTOF-MS) represents a universal tool for microscale enzyme assay, because capillary can be used not only as separation tool but also as reaction chamber. Method for reaction mixture mixing based on diffusion provides mixture with uniform concentration of reactants. In this contribution on-line CE-QTOF-MS method for study of diclofenac metabolism mediated by cytochrome P450 2C9 isoenzyme or human liver microsomes is presented. Michaelis constants were determined. |
