Description In 1993, protein p53 was hailed as the "Molecule of the year" by Science journal. It is not surprising that nowadays the protein p53 is the most studied object in molecular oncology area. Tetramers of the protein are made at cell stress. Tetramers subsequently bind to DNA and here they regulate proteins synthesis, thereby control reparation process. Structural form of the protein has significant influence on bond between p53 and damaged DNA. Three protein forms (native, denatured and aggregated) were studied by flow injection analysis with electrochemical detection (FIA-ED). The very sensitive method for native p53 form determination was developed; detection limit (LOD) 45.8 amol. FIA-ED technique was applied on study of protein p53 structural changes (formation of denatured and aggregated form). Denatured protein gives higher electrochemical response (protein structure release) and aggregated a smaller one (protein folding) in comparison with native form. Od strany: 120
