AnnotationA particular feature of chromosome ends is their mode of maintenance by the specific nucleoprotein complex of telomerase. Telomerase also participates in healing of chromosome breaks by adding telomeric sequences de novo. Telomerase in plant cells, in contrast to human cells, shows reversible down- and up-regulation depending on cell proliferation status. Therefore, plant telomerase is suitable target for research of structure-directed processes in regulation of telomerase activity. This project will deal with the fundamental questions of structure, function and regulation of telomerase in plant cells using model systems of Arabidopsis, Solanaceae and Asparagales. We will develop a system to identify interaction partners of Arabidopsis telomerase. This study will be accompanied by structural study of plant TERT subunit in detail. Further, we will test our hypothesis about possible scenario of telomere-telomerase evolution in plants using model plants of Solanaceae.
