Popis Nuclear location of subtelomeric and centromeric DNA sequences of chromosome 8, 9 and 13 was investigated in 3D fixed human G0 and stimulated T-lymphocytes using repeated dual colour FISH. In the first hybridisation, DNA probes for p and q arms labelled by different fluorochromes were used. The centromere and the chromosome territory of the same chromosome were visualised in the second hybridisation on the same nuclei. It was found for all investigated chromosomes that contrary to centromeres located in the proximity of the nuclear membrane, the telomeres adopt more central position. These results indicate a hairpin like structure of chromosomes in the interphase nucleus. The results of this study provide information not only on the topography of subtelomeres but also images of the spatial arrangement and orientation of interphase chromosomes in cycling and non-cycling cell nuclei.
