Description Alterations in glycan composition are closely linked to the progression of various diseases, making glycans valuable biomarkers. However, their analytical detection is challenging due to the lack of inherent chromophores. To overcome this, glycans must be dervatized with fluorescent tags. This study focuses on developing a new series of fluorescent markers designed for covalent glycan labeling, offering rapid reaction kinetics and high quantum yields suitable for chromatographic analysis via HPLC or CE. The fluorophore series comprises five compounds, each differentiated by specific substitution on a phenyl ring (methyl, methoxy, fluorine), which modulate their physicochemical properties. The core fluorescent scaffold is based on a pyrroloindolizine structure. Each compound is synthesized in two functional forms: one with a hydrazide group for neutral glycan labeling, and another with an N-hydroxysuccinimide ester for aminoglycan labeling. The prepared structures will be characterized in cooperation with the Institute of Analytical Chemistry of the Academy of Sciences of the Czech Republic. The practical application of these novel fluorophores will be demonstrated through glycoprotein profiling in breast cancer research.
