Description Hemocytes belong to the basic elements of invertebrate immune system. Increased reactive oxygen metabolites (ROM) generation was found in activated molluscan hemocytes (LAMBERT & NICOLAS, 1998; ORDAS et al. 2000), and therefore ROM are considered to be a significant microbicidal factor of invertebrate hemocytes similarly as of vertebrate phagocytes. The aim of this study was to analyse the ROM production of silkworm (Bombyx mori) hemocytes using various methods. Hemocytes were isolated from hemolymph of polyvoltine hybrid NO2CO2. Spontaneous chemiluminescence (CL) of hemocytes was at the background level. In following experiments hemocytes were activated by phorbol myristate acetate, zymosan particles or calcium ionophore A23187. The increase in CL was observed neither in this set of experiments. ROM production was also observed using spectrophotometric (NBT test, cytochrome c assay) and flow cytometric (dihydrorhodamine and hydroethidine staining) analyses. Again, no ROM production was observed. Similar results with CL method are also referred by other authors (e.g. MAZET et al., 1994) for other Lepidoptera species. In conclusion, no microbicidal activity dependent on oxygen was found in silkworm hemocytes. It is likely that insect hemocytes react differently in comparison with other invertebrate hemocytes and mammalian phagocytes and do not employ ROM to destroy invading microorganisms.
