Effect of ion concentration changes in t-tubules on membrane currents and intracellular Ca2+ transient in a model of human ventricular cardiomyocyte

Popis The transverse (t-) tubular system is formed by invaginations of the surface membrane that brings excitation deep inside the cell. In our previous study we formulated a mathematical model of human ventricular cell that included the t-tubular system. In the present study we improved the model by introducing description of new details based on available experimental data related to detailed morphology and properties of ionic currents in human ventricular myocytes. The simulations suggest that if the t-tubular fraction of Ca2+ channels (L-type) is high (> 60%) and the t-tubular fraction of sarcolemmal Ca2+ pump is low (< 30%) the activity induced transient reductions of Ca2+ concentration in human t-tubules might significantly lower Ca2+ entry into the cell and thus reduce the intracellular Ca2+ transients that control the strength of cellular contraction.
