Popis Successful application of matrix-assisted laser desorption/ionization time-of-flight (MALDI TOF) mass spectrometry imaging (MSI) at biomarker discovery, drug and metabolites distribution analysis and neural network studies provided this technique with an influential position among imaging techniques. Using label-free approach, MSI is able to detect and localize a wide range of ionizable substances in biological tissues. A disadvantage of this technique is speed of the analysis. Since the very beginning, a single acquisition of a high-resolution MS image could take even days. Recent commercial mass spectrometers employing high-repetition rate solid-state desorption lasers significantly improved MALDI MSI throughput, but the analysis time is still in the order of hours. Here we introduce a novel sampling approach taking advantage of incorporation of a high-speed precision scanning mirror redirecting the desorption laser beam across the imaged sample area. Thus the scanning mirror partially substitutes a few order slower sample holder stages translation. A laboratory-built instrument equipped with 2 kHz 355 nm Nd:YAG laser and the fast scanning mirror recorded 10 000 pixel MS image with 100 laser shots per pixel in 13 minutes, an order of magnitude faster compared to a recent commercial instrument with 1 kHz laser.
