Popis Hydrolysis of nucleic acids mediated by artificial nucleases is very important goal in molecular biology, biotechnology and medicine. Some metal and lanthanide ions and their soluble or insoluble complexes (immobilized on solid particles) are attractive as catalysts for DNA cleavage. The aim of this work was to use of neodymium ions for cleavage of plasmid DNAs. Plasmid DNAs originated from strains of Staphylococcus aureus isolated from clinical and food samples were treated by neodymium ions (Nd3+). Plasmid pUC19 was used as model molecule and linearized pUC19/EcoRI was used as the control of the presence of linear form. Cleavage of plasmid DNA was carried out at different temperatures in HEPES (0.05M) buffer. The reaction time was up to 4 hours. The reaction was stopped with 0.5 M EDTA (pH 8.0). Plasmid DNA cleavage efficiency was evaluated by convert the supercoiled circular (ccc) form of plasmid DNA into nicked circular form (oc) and linear (lin) one and was monitored on the gel according to the shift of their mobilities.
