Concurrent infection of porcine monocyte-derived macrophages with PRRSV and Haemophilus parasuis

Popis Porcine reproductive and respiratory syndrome (PRRS) is an important swine disease causing severe economic losses worldwide and can predispose pigs to infection by bacteria such as Haemophilus parasuis. This study was aimed on immune response of porcine monocyte-derived macrophages (MDMF) to simultaneous infection with PRRSV and H. parasuis in vitro. Expression of immunologically-relevant molecules (measured by qRT-PCR) and antimicrobial characteristics such as respiratory burst (measured by luminometry) were tested. MDMFs were sensitive to PRRSV infection and its mortality after 24 hours with PRRSV was high (85%). Concurrent infection with PRRSV and H. parasuis induced lower expression of mRNA for pro-inflammatory cytokines IL-1beta and IL-8 when compared to control. Anti-inflammatory effect of dual infection was also observed on higher expression of mRNA for IL-10. On the contrary, simultaneous infection of MDMF with PRRSV and H. parasuis induced higher expression of mRNA for pro-inflammatory cytokine TNF-alpha. Increased production of reactive oxygen species (ROS) was detected in MDMFs infected with PRRSV in comparison with non-infected MDMFs. On the other hand, simultaneously infected MDMFs decreased production of ROS. Obtained data showed that concurrent infection of MDMFs with PRRSV and H. parasuis elicits anti-inflammatory immune response. PRRSV infection seems to be responsible for elevated levels of ROS which might be one of the causes of high mortality of MDMFs.
