| Popis | The functional sample consists of HeLa cells prepared by high-pressure freezing and freeze substitution, followed by embedding in Durcupan epoxy resin, using the established protocol for cryo-fixation and resin embedding of biological samples. After polymerisation, the resin block is introduced into a focused ion beam (FIB-SEM) instrument. A shallow wedge is milled into the specimen under a well-defined 10° angle, producing a large, planar imaging surface with controlled geometry. To enable precise correlation between imaging modalities, fiducial marks are created by FIB at the edges of the wedge face. Subsequently, the wedge surface is imaged in the SEM using both secondary electron (SE) and back-scattered electron (BSE) detection modes. Directly on the same wedge face, AFM measurements are performed without further sample manipulation. The previously milled fiducial marks serve as reference features for high-precision spatial alignment between SEM and AFM datasets. |
