Bioassay-guided Isolation of Compounds with Anti-inflammatory Activity from Glycyrrhiza Species

| Popis | Glycyrrhiza species have long been used in folk medicine worldwide. In Western pharmacopoeias, Glycyrrhiza is mainly regarded as an expectorant, whereas in Asian traditional medicines it is widely used in anti-inflammatory formulations, drawing scientific attention to its pharmacologically relevant constituents. Glycyrrhiza species extracts are therefore the best candidates for isolation of compounds with anti-inflammatory activity. Ethanolic extracts of six Glycyrrhiza species (G. glabra, G. uralensis, G. echinata, G. foetida, G. pallidiflora, and G. yunnanensis) were sequentially fractionated (n-hexane, chloroform, and ethyl acetate) and analyzed by HPLC-DAD for phenolic content. CHCl3-soluble and EtOAc-soluble extracts were assayed in NF-?B inhibition tests performed on THP-1 cell line. In accordance with NF-?B inhibition results, extract from G. uralensis was prepared on larger scale and separated by RP-C18 chromatography. The most prospective fractions were selected via the NF-?B inhibition assay and subjected to isolation of compounds by semi-preparative HPLC. The chemical structures were elucidated by evaluation of their NMR and HRMS spectra. Finally, pure compounds were tested for NF-?B inhibition. CHCl3-soluble extracts from all Glycyrrhiza species extracts showed significant inhibitory activity on NF-?B signaling pathway, while EtOAc-soluble extracts did not show any significant activity. G. uralensis CHCl3-soluble extract exerted the most pronounced effects, hence selected for further investigation. As a result, three prenylated isoflavanones and six prenylated isoflavones were selectively isolated using bioassay guidance. Experimental investigation has shown a considerable potential of Glycyrrhiza species in the search for anti-inflammatory drugs, with G. uralensis exerting the most pronounced effects. Diprenylated isoflavonoids isolated by bioassay-guided fractionation may be considered for further investigation as potent NF-?B inhibitors with potential for drug candidates. In addition, our study supports the hypothesis that the presence of prenyl moieties is essential for enhancing the anti-inflammatory activity of phenolics. |
