Intracellular Fate of a Dual-Fluorescent Hydrophobic Ion Pair in Lipid-Based Nanocarriers

| Popis | The intracellular delivery of hydrophilic compounds re-mains challenging due to their poor membrane permeability and low encapsulation efficiency in lipid-based nanocarriers [1]. Hydrophobic ion pairing (HIP) has emerged as an effec-tive strategy to enhance the lipophilicity of hydrophilic molecules by forming ion pairs with oppositely charged counterions, thereby enabling their incorporation into lipid-based systems. In this study, a dual-fluorescent HIP was developed by pairing the hydrophilic dye Cascade BlueTM hydrazide with the lipophilic probe DiA, yielding a highly lipophilic complex suitable for encapsulation into lipid-based nanocarriers. This dual-fluorescent system allowed simultaneous visualization of both components to investi-gate their intracellular fate after delivery. The HIP was in-corporated into self-emulsifying drug delivery systems (SEDDS), nanoemulsions, and liposomes, enabling a com-parative assessment of how carrier composition influences cellular uptake, localization, and endosomal escape. |
