Lytic cycle dynamics of the Mammaliicoccus sciuri podovirus in different hosts

Popis Objective: M. sciuri is an emerging opportunistic pathogen colonizing various environments, including animals, humans, and plants. Hence, it is important to seek ways to eradicate it, including phage therapy. Here, we analyze the lytic podovirus S10 life cycle in two different host strains. Methods: We assess phage adsorption in the presence of various cations and the replication via a one-step growth curve on two different host strains. Transcription of phage S10 in these strains was assessed. Results: In all tested conditions, approx. 90% virions were adsorbed within 20 minutes. The phage replicated efficiently in one host strain, whereas in another it depended on the strain used for previous propagation. Phage transcriptomics revealed relative host-dependent differences in individual gene transcription, and a 4 orders-of-magnitude difference in total transcription. Conclusion: Phage S10 adsorption is divalent cation-independent. For one of the strains, the efficiency of phage replication depended on which strain was used for the previous round of replication. Possibly because the phage requires prior epigenetic adaptation. Therefore, a currently unknown anti-phage mechanism that silences transcription and an epigenetic system of the phage that overcomes it are hypothesized.
