| Popis | Objective: Redirection of bacteriophage capsid size is a common strategy used by phage satellites to accommodate their genome and gain an advantage over helper phages. We investigated how capsid-remodeling proteins affect the capacity of small-headed phage particles. Methods: Capsid-remodeling proteins from different Staphylococcus species were tested for their effect on virion capacity. Phage capsids were reconstructed by cryo-EM, and packaged genome lengths were analyzed using Oxford Nanopore sequencing. Results: Remodeling proteins of different origins were tested on a panel of Staphylococcus aureus and Staphylococcus epidermidis phages for their ability to induce small-capsid formation. Structural analyses revealed different retention of remodeling proteins, resulting in altered DNA capacity of small-headed virions. Conclusion: Variation in capsid-remodeling proteins modulates their interaction with helper phages, leading to differences in the amount of DNA packaged into small-headed virions. This altered capacity constrains the size of genophores and determines the genome length compatible with efficient transduction of phage satellites by small-headed virions.
