Regulation of many genes in denitrification bacteria is mediated by members of the E. coli FNR class of transcription activators. Disruption of their iron-sulfur centers upon direct reaction with oxygen diminishes the ability to form a dimer and bind DNA . However, there are observations suggesting that oxygen may also act indirectly by decreasing the degree of reduction of intracellular redox active components and affecting the cytoplasmic concentration of Fe(II). Three complementary ways to explore thi s hypothesis are proposed. (1) Gene expression at the protein level (2D-gel electrophoresis protein database, individual hemoproteins and respiratory enzymes), the content of individual hemes and FNR-dependent promoter activity can be assayed to compare the responses of the oxically grown cell culture to intracellular redox shifts due to limited aeration, the inhibition of respiratory enzymes or the presence of a thiol compound.
