Description We developed a Raman spectroscopy method for evaluating the DS of HA modifi ed by saturated acyl side-chains. The DS is calculated from a band intensity ratio using independent calibration on palmitoyl-HA oligosaccharides with known DS. The Raman results agree very well with data obtained by the established GC method, suggesting that it can be used for routine sample analysis. Advantages of the Raman method: • Samples are measured directly in the solid state, simplifying sample preparation (no dissolution, hydrolysis, etc.). • Can be used for non-destructive analysis of application forms. • Low cost (only instrument and analyst time). • Short analysis time (< 90 min from sample delivery to data with RSD). Disadvantage: can not diff erentiate free fatty acids.
