Description Spirochetes from Borrelia burgdorferi sensu lato are originators of serious infectious diseases Lyme borreliosis. The main physiological properties of these spirochetes are variable antigenic equipment enabling adaptation to a wide spectrum of hosts. Lipoprotein OspC is one of the most important antigens using during the transmission to the host. The aim of our study was to develop recombinant forms of OspC relevant to the Czech Republic, against these antigens prepare a set of mouse monoclonal antibodies and identify binding epitopes recognized by these antibodies. From the tested hybridomas was selected 9 lines based on the ability to bind to both recombinant and natural forms of OspC (B. garinii, B. burgdorferi sensu stricto, B. valaisiana and B.afzelii). These antibodies were purified using affinity chromatography (protein G), and further characterized. In the three lines with the highest affinity (3F4, 2D1 and 2A9) were identified epitope by analyzing the peptide of cleaved antigen bound to the immobilized antibody using mass spectrometry.
