In-cell NMR spectroscopy is the only analytical method that enables the structural characterization of proteins and their interactions in living cells at atomic resolution. It has recently emerged as a unique tool for addressing various research questions related to human physiology and drug development. Its applications include target structure validation, drugtarget engagement assessment, and evaluation of drug-target affinity under native-like conditions. However, in-cell NMR spectroscopy in human cells has been limited to asynchronous single-cell(-type) cultures. Unfortunately, these cultures do not reliably recapitulate the key functional properties that cells exhibit when part of a tissue. Additionally, many cell types cannot be maintained in traditional culture conditions but can only be propagated when differentiated in situ from progenitor (pluripotent stem) cells as part of a human tissue-like environment. Reliance on single-cell(-type) cultures represents a major limitation of current in-cell NMR applications in drug development and basic research.
