Description Experimental study of the nature of heritable arrhythmias is possible in a variety of ways, from in vitro analysis of changes in the properties of ion channels transfected in a cell line to in vivo investigation in an animal model. In recent years, the model of patient-specific cardiomyocytes differentiated from human induced pluripotent cells (hiPSCCM) has become a promising and increasingly used model for the study of inherited arrhythmias. Its advantages are indisputable. To obtain, with a completely minimal burden on the patient, his or her own cardiomyocytes and to be able to subject them, thanks to the infinite possibility of hiPSC proliferation, to virtually any extensive series of in vitro assays is astonishing and is not made possible by any of the other possible models. On the other hand, it is a technically, time and cost intensive methodology. A fundamental problem is the immaturity of the obtained hiPSC-CMs. None of the differentiation protocols currently used allows to achieve sufficient maturity comparable to adult cardiomyocytes. This limits the transfer of the obtained knowledge to clinical practice.
