Description We have shown that the whole spectrum of cytokinins and their derivatives does occur in tobacco and wheat chloroplasts (Benková et al., Plant Physiol. 121: 245-251, 1999). We have further compared cytokinin levels in chloroplasts isolated from control tobacco plants (SR1) and plants carrying the Zmp-60.1 gene, coding for a beta-glucosidase and being naturally targeted into chloroplasts. Preliminary experiments did not show any significant difference between chloroplasts from control and transgenic plants. Further experiments are under progress. To stabilize chloroplasts we developed a technique of chloroplast immobilization in Ca-alginate. Chloroplasts remain metabolically active for at least 20 h.
