Influence of micromanipulations on the cytoskeleton of human cells in vitro

Micromanipulation techniques have an increasing importance within the range of methods of the contemporary cell biology and reproductive technologies. The aim of the project is a detailed study of the behaviour of cytoskeletal structures in cells in vitr o subjected to of mechanical damage by micromanipulations. Various types of diploid and established human cell lines bearing different morphological markers will be used as model objects. The dynamic changes of cytoskeletal components (microtubules, micr ofilaments and intermediate filaments) will be followed as well as the influence of mechanical damage on mitotic divisions and intracellular transport. In addition, the role of the cytoskeleton in reparative processes after micromanipulation invasion wil l be studied. Immunofluorescence detection of the cytoskeletal proteins using of the monoclonal antibodies, transmission and scanning electron microscopy will be the main experimental approaches.
