Description Borrelia burgdorferi sensu lato is a gram-negative spirochete causing multisystem prolonged disease - Lyme borreliosis that problem-free antibiotic treatment significantly depends on early diagnostics and identification of pathogen. No reliable vaccination against borreliosis was developed for human medicine to this date. The most common global vector of transition is tick (Ixodes ricinus) and its developmental forms, although possibility of infection by a different blood sucking anthropod is not excluded. In our study which was realized from 2008 till October 2012 we analyzed 944 samples mostly of ticks for occurrence of B. burgdorferi s. l. The isolation of DNA was performed using UltraClean BloodSpin DNA Kit (MoBio). For identification of pathogen was used a high sensitive EliGene Borrelia LC (Elisabeth Pharmacon) kit which allows detecting 1–10 Borrelia flagellin DNA in 5 ul of clinic sample by using RealTime PCR systems on the instruments LightCycler 2,0, LightCyler 480, LightCycler Nano, RotorGene and ohters. Average infection rate was 23 % during those four years. Among regions with the highest portion of infected ticks belonged to Jihočeský and Pardubický region. From analysis of our data follows the occurrence of infected ticks which correlates well with annual and monthly weather in the Czech Republic (rain, temperature). From sequence analysis of intergenic spacer (IGS) of positive samples follows the highest occurrence of B. afzelii then B. garinii and the other species. Among others the sequence analysis confirmed robustion of our method of detection and its possible application for early diagnostic and subsequent suitable selected antibiotic treatment.
