| Description | Sulfate-reducing bacteria (SRB) represent key anaerobic microorganisms with a significant impact on animal physiology as well as on industrial corrosion, while often competing with methanogens in the environment. This study focuses on the optimization of cultivation conditions for SRB and their subsequent isolation from mixed cultures. In the first part of the experimental work, the growth of the strain Desulfovibrio desulfuricans DSMZ 9104 was monitored, and a positive effect of enriching media with Wolfe’s vitamins and minerals or rumen fluid on biomass increase was demonstrated. To evaluate the effect of additives on growth, the Bradford method was used, as standard optical density measurements were distorted by the formation of sulfide precipitates. In the next part of the study, pure cultures were isolated from mixed samples using techniques based on the “agar shake dilution” and “Hungate roll tube” methods. Identification of the obtained isolates was carried out based on cultivation characteristics and molecular detection of the functional gene aprA. The main outcome of this work is the successful isolation and characterization of a pure strain of Nitratidesulfovibrio vulgaris. |
