Description Human leukemic promonocytes U937 can be induced to differentiate to both monocytes/macrophages using phorbol ester TPA and granulovytes using retinoic acid (RA). The v-myb oncogene of avian myeloblastosis virus codes for protein that transforms myelomonocytic cells in vitro and in vivo. In order to investigate the function of this protein, we transferred and expressed the v-myb cDNA in U937 cells. The v-Myb protein suppressed both TPA-induced monocytic- and RA-induced granulocytic differentiation of U937 cells. In addition, v-Myb increased resistance of U937 cells to inducer of apoptosis, camptothecin. This suggests that transforming function of the v-Myb oncoprotein results from its capability to suppress both differentiation and programmed cell death of progenitor myeloid cells.
