The role of cytoskeleton and cell wall in the cell polarization

The role of cytoskeleton (actin, microtubules and membrane skeleton) in the process of the cell polarity will be studied on the model of budding yeast Saccharomyces cerevisiae and the fission yeast Schizosaccharomyces japonicus var. versatilis. Mating process, protoplasts regeneration and osmotic stress will be studied as processes changing cell polarity. Cdc mutants showing defect in the cell polarization (cdc24,42), mutant in Kar 3p controlling karyogamy, and mutants in high osmolarity glycerol pat hway (hog1 and pbs2) as well as microtubule and actin inhibitors will serve as an experimental approaches. Fluorescence microscope techniques, including indirect immunofluorescence, and electron microscope techniques will be applied in the study of chang es in cytoskeleton and cell structure.
