Description The present invention provides an absolute quantification method of miRNA expression, being preferably selected from miR-34a and/or miR-150, wherein miRNA expression, being preferably selected from miR-34a and/or miR-150, along with the expression of endogenous check, which is a small nucleic RNA or permanently expressed miRNA are determined in a biological sample being withdrawn from a patient body. At the same time, there is determined expression of synthetic standards of miRNA, being preferably selected from miR-34a and/or miR-150 and endogenous check of known number of molecules. The invention further provides the use of the above-indicated quantification method for in vitro diagnostics of B-cell malignancies, prognostics of B-cell malignancies and prediction of therapeutic response of patients with B-cell malignancies, especially CLL, identification of TP53 abnormalities.
