| Description | Plasminogen was included in the research of new thrombolytics and their combinations. It was shown that the combination of alteplase with plasminogen led to a statistically significant improvement in thrombolysis compared to alteplase alone. Determination of the concentration of plasminogen and plasmin allows the calculation of the PLASMINOGEN/PLAZMIN ratio, which reflects the level of plasminemia. It can thus predict the effectiveness of thrombolytic drug therapy and the associated risk of hemorrhagic complications after thrombolytic therapy. HPLC could offer faster analysis, lower operating costs and better repeatability. The main result of the work is the optimization of conditions for the separation of plasminogen and albumin using RP-HPLC. It turned out that a higher temperature is more advantageous due to partial denaturation and exposure of hydrophobic amino acids for stronger interaction with the stationary phase. The C8 column provides better separation than C4, because it allows stronger interactions with hydrophobic regions of proteins and accentuates the difference in retention between analytes. A shorter gradient is more effective for separation, because it leads to a faster change in elution strength and less diffuse broadening of peaks. The result is a method with the potential for rapid and reproducible analysis of fibrinolytic system proteins. HPLC may be used as a practical alternative for monitoring markers of thrombolytic therapy. |
