Description Backgrounds: In the last decade various new technologies in medicine and molecular biology have been developed, among others cDNA microarray technology. cDNA microarray technology enables comparison of expression profiles of tested samples and it is convenient for diagnostics of diseases on genes level. Design and Subject: The aim of this study was comparison of gene expression of colorectal carcinoma and colon epithelium and detection of differences in expression profiles of patients with regional metastases and non-metastatic patients. The study was performed on the group of 12 patients with colorectal carcinoma. Methods and Results: 1.7K Human cDNA microarrays containing 1700 fragments of human cDNA were used for determination of expression profiles of patients. We have found 22 genes with altered expression in colorectal carcinoma as compared with normal tissue. Using hierarchical clustering, patients were divided into two groups according to occurrence of regional metastases: patients with regional metastases and non-metastatic patients. These two groups differed in expression of 6 genes. Conclusion: cDNA microarray technology, based on the detection of changes in the expression profiles, provides easier, quicker and more effective diagnostics of various diseases. In future, genes found by cDNA microarrays in our experiments, could constitute a part of the newly developed diagnostics colon cancer chips.
