Popis Benzo[c]phenanthridine alkaloid sanguilutine (present in plant Sanguinaria canadensis) was extracted and separated from the plant material (dried roots). Interaction of the positively charged iminium form of alkaloid with double stranded DNA is reported in this article. Significant changes in fluorescence properties in the presence or absence of various oligonucleotides have allowed us to study this system by means of luminescence spectroscopy. Results have shown that interaction with different double stranded oligonucleotides is not specific to A-T or G-C base pairs and also no preference of homogeneous or heterogeneous base composition of strands was found. Association constants (log K) were calculated to be in the range of 5.11 to 6.16. The response of luminescence intensity at 610 nm to low concentrations of double stranded DNA was found to be linear and can be potentially used for fluorometric quantification of DNA. The limit of detection was estimated to be 120 ng.ml-1 of DNA (calculated by 3 sigma method).
