Popis Nitric oxide is produced by the nitrite reducting nitrite reductase and further consumed by nitric oxide reductase. Time course of nitric oxide concentration was monitored amperometrically by a Clark type electrode and simultaneously by using direct inlet mass spectrometry. pH considerably affected the accumulation of NO. Using ascorbate and N,N,N,N, tetramethyl p phenylendiamine (TMPD) as a source of electrons, oscillations of nitric oxide concentration were observed in the pH range of 6 6,4. Nitric oxide did not accumulate at higher pH values, while the accumulation proceeded at lower pH. This coresponds well with pH dependences of the activities of both involved enzymes (pH optimum of nitritreductase = 6.2 and pH optimum of NO reductase = 7.5). Oscillations of nitric oxide concentration can be elicited by a partial inhibition of nitric oxide reductase with the uncoupler carbonyl cyanide m chlorophenylhydrazone (CCCP). Parameters of NO oscillations were found to be influenced by some factors: concentrations of electron donor TMPD or biomass and reaction temperature.
