Popis The potential human pathogen M. pachydermatis was studied to identify cytoskeleton as potential therapeutic target for antifungal agents. Light and transmission electron microscopy (TEM) confirmed the small cells (2,0-3,0 x 4,0-5,0 mm) and few intracellular structures. TEM confirmed the presence of thick multilayered cell wall that appeared to be a barrier for fluorescent antibodies. The higher concentration of detergent and lytical wall enzymes, the prolonged time of their and fluorescent antibodies applications seem necessary prerequisites to detect cytoskeleton. Preliminary results showed poor cytoskeletal structures. This report confirmed multilayered cell wall, the deep and helicoidal invaginations of plasma membrane and few intracellular structures. Cytoskeleton will be further studied by fluorescence and immunofluorescence and by TEM. The research was supported by grant No. 310/00/0391 of the Grant Agency of Czech Republic.
